Guitar into interface, interface out into computer and monitors, Petrucci standalone on laptop… All I can hear is my raw guitar sound out of the speakers… No Petrucci at all.
I’m sure I’m not the first one, total newbie btw… What am I doing wrong??

Go into the plugin standalone settings and configure your inputs/outputs to the applicable interface.
OK, my settings are
Audio device :windows low latency mode
(if I put it on ASIO, all sound stops )
Audio output :USB audio device
Audio input device :USB audio device
Sample rate 48000 hz
Audio buffer 96 samples 2.0 ms
OK, the problem I have NOW is, I can hear the distorted guitar sound, but the latency is over the top and it has some ringing (kinda feedback sounding, like a reverb from hell) noise when I abruptly mute the strings…
Could this possibly be my interfaces fault?
What are your thoughts?

You might want to remove your number to prevent people SPAMing you etc.
This is most definitely an issue with your audio interface and possibly the configuration(s) within. Make sure your interface drivers are up to date and you definitely want to be using the ASIO driver that comes with your interface (PC only). Once you have that working, tweak your sample size until you have the best performance.
